CYCRAME GENERIC:
CYCLOPHOSPHAMIDE

CLASSIFICATION:
Alkylating agent

MECHANISM OF ACTION:
Active metabolites interfere with growth of rapidly proliferating malignant cells. Polyfunctional alkylating agent may cross-link DNA, interfering with DNA replication and transcription of RNA, disrupting nucleic acid function. Also has potent immunosuppressive activity. The drug exhibits phosphorylating properties that also enhance its cytotoxicity.

MECHANISM OF ACTION:
Daunorubicin has antimitotic and cytotoxic activity through a number of proposed mechanisms of action: Daunorubicin forms complexes with DNA by intercalation between base pairs, and it inhibits topoisomerase II activity by stabilizing the DNA-topoisomerase II complex, preventing the religation portion of the ligation-religation reaction that topoisomerase II catalyzes.

STORAGE:
Store unopened vials in refrigerator, 2° to 8°C (36° to 46°F). Store prepared solution for infusion at room temperature, 15° to 30°C (59° to 86°F) for up to 24 hours. Contains no preservative. Discard unused portion. Protect from light.

MECHANISM OF ACTION:
As a pyrimidine analogue, it is transformed inside the cell into different cytotoxic metabolites which are then incorporated into DNA and RNA, finally inducing cell cycle arrest and apoptosis by inhibiting the cell's ability to synthesize DNA. It is an S-phase specific drug and only active during certain cell cycles.

MECHANISM OF ACTION:
Vincristine prevents the formation of spindle fibers during metaphase of mitosis. It binds to tubulin, the protein that makes up the spindle fiber microtubules, and prevents metaphase of mitosis.

CLASSIFICATION:
BETA LACTUM ANTIBIOTIC

MECHANISM OF ACTION:
Meropenem is bactericidal except against Listeria monocytogenes where it is bacteriostatic. It inhibits bacterial wall synthesis like other beta-lactam antibiotics. In contrast to other beta-lactams, it is highly resistant to degradation by beta-lactamases or cephalosporinases. Resistance generally arises due to mutations in penicillin binding proteins, production of metallo-beta-lactamases, or resistance to diffusion across the bacterial outer membrane. Unlike imipenem, it is stable to dehydropeptidase-1 and can therefore be given without cilastatin.

MECHANISM OF ACTION:
Hydroxyurea is thought to be cell cycle-specific for the S phase of cell division. The exact mechanism of antineoplastic activity is unknown but is thought to involve interference with synthesis of DNA, with no effect on synthesis of RNA or protein.

MECHANISM OF ACTION:
Cisplatin is similar to the bifunctional alkylating agents. It covalently binds to DNA and disrupts DNA function. After cisplatin enters the cells, the chloride ligands are replaced by water molecules. This reaction results in the formation of positively charged platinum complexes that react with the nucleophilic sites on DNA. These platinum complexes covalently bind to DNA bases using intra-strand and inter-strand cross-links creating cisplatin-DNA adducts thus preventing DNA, RNA and protein synthesis. This action is cell cycle phase-nonspecific. Cisplatin has immunosuppressive, radiosensitizing, & antimicrobial properties.
